SRX275929: Survey of transcriptomes Brevibacillus borstelensis in response to low and high temperature shock

Design: Bacterial culture of Brevibacillus borstelensis was grown overnight in NB media and RNA was isolated from cells heat shocked at 55 degree cellsius for 5 minutes. Ribodepleted total RNA(mRNA) was isolated and then fragmented using RNase III enzyme. Fragmented mRNA was hybridized with adapters followed by ligation. Reverse transcription was carried out to obtain cDNA and then size selected libraries were amplified. Finally transcriptome library was quantified and qualified using bioanalyzer.

Study: Brevibacillus borstelensis Transcriptome or Gene expression

To identify and understand the role of stress-relevant genes responsible for the survival mechanism of the thermophiles in a wide range of environment, whole transcriptome analysis of Brevibacillus borstelensis was performed de novo by Illumina Hi Seq 2000 sequencing technology. The gene expression profile of B. borstelensis was analyzed after the exposure of the bacterium to low temperature treatment (20°C for 30 min) and high temperature treatment (55°C for 5 min and 10 min). The total number of genes that revealed significant similarity to known functional protein-coding genes were annotated using the GO and KEGG pathway databases. Differential profiling of the genes expressed in the control (50°C) and treated samples revealed the significantly up regulated and down regulated genes. This brief survey of the transcripts obtained in response to low and high temperature shock underline the survival strategy of a thermophilic bacterium under different temperature parameters that is further helpful to generate vast genetic information associated with the bacteria.
